Polarimeter is mainly used to determine the rotation angle and international standard sugar content. The measurement of the rotation angle can ensure the quality and safety of the product by measuring the specific optical rotation of pharmaceuticals, perfume chemicals and the like. Polarimeters can be used to determine the rotation angle or optical rotation of a sample aqueous solution. Specific optical rotation is an intrinsic property of a substance at a specific temperature and wavelength and can be calculated from the length of the glare tube, rotation angle, and concentration. The international standard sugar content is used to determine the purity of the raw sugar during the sugar production process. The sample is measured using the “international standard sugar content†function on the polarimeter. The measured value is the international standard sugar content of the sample. Polarimeters used in the pharmaceutical industry for the determination of the optical rotation of pharmaceutical raw materials and finished products, the measurement of components in the fine chemical industry or the composition of perfumes and secondary optically active substances, the measurement of the purity of sugar in the food industry or the quality of invert sugar, and the ratio of optically active compounds Measurement of curl or content.
Polarimeters are the more common and important laboratory testing instruments in the laboratory. In order to allow customers to better use and maintain the polarimeter, Guangzhou Aibei Scientific Instrument Co., Ltd. will tell you about the use and maintenance techniques of the automatic polarimeter, reduce misoperation and instrument failure, and extend the life of the instrument.
A. Polarimeter construction components and accessories Consumables: 1. Light source 2. Frosted glass 3. Condenser 4. Color filter 5. Polarizer 6. Half-wave plate 7. Test tube 8. Analysis lens 9. Object, eyepiece group 10. Focusing Hand wheel 11. Magnifying glass for reading 12. Dial and cursor 13. Turning the hand wheel when the test tube is placed in the test cell After the test solution is stored in the test tube, since the solution is optically active, the plane of polarized light is rotated by an angle, zero degrees. The field of view changes, turning the analyzer at a certain angle, and the field of view with the same brightness can appear again. This rotation angle is the optical rotation of the solution. After determining the optical rotation of the solution, the specific rotation of the material can be determined. Based on the specific rotation, the purity and content of the substance can be determined.
Second, the use of polarimeter:
(1) Connect the polarimeter to 220V AC power. Turn on the power switch, and after about 5 minutes, the sodium lamp will glow normally and you can start working.
(2) Check whether the zero position of the polarimeter is accurate. That is, when the polarimeter is not placed in a test tube or placed in a test tube filled with distilled water, observe whether the brightness of the field of view is the same at zero degree. If there is an inconsistency, there is a zero error, which should be subtracted or added to the measurement reading. Or relax the cover plate with four screws on the back, and slightly adjust the dial cover to correct it (correction error of only about 0.5°, which should be sent to the manufacturer for repair).
(3) Select the appropriate length of test tube, fill the liquid to be tested, install the rubber band, and screw on the nut until it does not leak water. Nuts should not be screwed too tight, otherwise the guard glass will cause stress and affect the correctness of the reading. Then dry the remaining solution on both ends of the test tube to avoid affecting the observation clarity and measurement accuracy.
(4) Determine optical rotation readings: Rotate the dial, the analyzer, and obtain the same brightness in the field of view, and then read from the dial. A positive reading is a right-handed substance, and a negative reading is a left-handed substance.
(5) The double-cursor reading method can be used to obtain the result according to the following formula: Q = (A + B) / 2 where: A and B are the two cursor window readings, respectively. If A = B, and the dial to any position is in accordance with the equation, then there is no eccentricity difference (usually before the factory polarimeter has been corrected), you can not use the item reading method. (6) Optical rotation and temperature are also related. For most materials, measured with λ = 5893 A° (sodium light), the optical rotation decreases by approximately 0.3% when the temperature is increased by 1°C. For more demanding measurements, it is best to perform at 20 °C ± 2 °C.
Third, the maintenance of polarimeter (1) Polarimeter should be placed in a dry and well-ventilated place, in order to avoid damp moldy.
(2) The continuous use of the polarimeter should not exceed 4 hours. If the use of a long time, the middle should be turned off for 10 to 15 minutes, until the sodium lamp cooling and then continue to use, or blow with a fan to reduce the degree of heating the lamp to avoid brightness and life expectancy.
(3) After the test tube is used, the solution should be poured out in time, washed with distilled water, and dried. All lenses must not be rubbed by hand and rubbed with a soft fleece.
(4) If there is any problem during the use of the polarimeter, contact the ATAGO (China) after sales engineer in time.
(5) When the polarimeter is deactivated, the plastic sleeve should be put on. When packing, it should be placed in the box in a fixed position and pressed tightly.
